Is it possible to run sdrconle with adalm pluto on TX and rtl sdr key for rx?

Not possible running one instance. You would need two instances of SDRC, one for the RTL and one for the Pluto. Then it could be done, yes, but frequency control would be totally independent of each other.
I'm not sure if anyone has tried to sync two instances of SDRC together via com ports. In theory I think that may be possible although pretty sure it would need two identities to keep the com port settings separate. Then of course you would need to sort out the antenna switching and PTT to mute the receiver. I guess the RX could be left to auto-mute. Goes without saying you'd need to be careful not to blow the RTL with excessive RF. I'm sure you already thought about this.

All for QO-100 sat, one instance for Tx ( pluto ) and another for rx ( rtl sdr ). And how all work.

I see that i have better rx with my rtl sdr key, and want you try it just for curiosity. And for the final compare the two rx system.
